Phase I: Immunogen design and Immunization (8-9 wks).
- Design and preparation of custom peptide immunogens or protein antigens.
- Site directed KLH/BSA conjugation for peptides.
- Multiple immunizations of Balb/c mice (n = 5).
- Test bleed antibody titering and qualification prior to the final boost and cell fusion in Phase II.
Phase II: Cell fusion and parental clone screening (3-wks).
- Approximately 6-8 weeks after initial immunization, mouse with the highest antisera titer will be given a final boost for cell fusion.
- Fused cell clones are screened by ELISA.
- Potential parental clones will be expanded into 24-well plates to harvest relatively large amount of tissue culture supernatant for further antibody screening (ELISA, WB or IHC).
- Shipment of up to 12 supernatants (0.5-1 ml/per parental clone).
- Customer must feedback within 1 week to ensure viability of clones.
Phase III: Monoclonal selection and cryogenic vial preservation (3-wks).
- Monoclonal selection of the best 1-3 clones will be performed (based on the customer’s choice).
- Subclone(s) with high specificity and antibody titer will be expanded and cryopreserved.
- 10 ml of supernatant and the cryopreserved vials (3 vials) from each clone will be shipped to customers.
- Customer must feedback within 1 week to ensure viability of clones.
Phase IV: Large scale antibody production (3-6wks) (optional)
- In vivo ascites production (20-40 ml of ascites each clone) or In vitro Bioreactor antibody production (up to 120 ml of supernatant each clone).
- Monoclonal antibody purification from ascites or culture supernatant using affinity chromatography. ELISA and SDS-PAGE will be performed to ensure the quality of the purified monoclonal antibody.
- AnaSpec provides both small and large scale antibody production with flexible cost adjustment.
IMPORTANT NOTE: AnaSpec will store the parental clones and subclones for 3 months only. Unless contacted by customer within the 3-month period, these clones will be discarded. Storage fee applies if customers choose to keep the clones at AnaSpec.
