As in the case of ß-Secretase Substrate 1, this FRET peptide is an internally quenched fluorescent b-Secretase substrate where the Val residue is replaced by Met. Upon cleavage by ß-Secretase, the Mca (7-Methoxycoumarin-4-acetic acid) fragment is liberated from the proximity quenching effect of Dnp (2,4-dinitrophenyl), Abs/Em=325/393 nm.
