Species Reactivity: Human, rat and mouse. In IHC, rabbit polyclonal anti-Beta-Actin (CT) antibody showed moderate to strong staining across a wide variety of human formalin-fixed tissues with minimal background staining. At least 22 different human formalin-fixed, paraffin archival tissues, and positive and negative tissues were scored and compared to the published literature on the expression and function of the gene.
Application: WB, IHC
Product Description: Rabbit polyclonal anti-beta actin antibody is supplied as an epitope affinity purified rabbit IgG, 50 µg in 250 µl (0.2 mg/ml) 1x PBS (pH 7.4) containing 0.1% BSA and 0.05% sodium azide.
Immunogen: Rabbit polyclonal anti-Actin antibody was raised against a 16 amino acid-peptide from near the carboxy terminus (CT) of human beta-actin (Genbank accession No. AAH02409).
Application Notes: The following concentration ranges are recommended starting points. Optimal working concentrations for specific application should be determined by the investigator.
ELISA for immunizing peptide: 1: 5,000-20,000 WB: 1: 500-1,000 IHC:1:100-200
Figure 1. IHC of beta-actin in human testis tissue labeled with rabbit polyclonal Anti-beta-Actin (CT) antibody (Cat# 54590). Beta-actin is shown as the precipitated red signal, and moderate staining was seen in myofibroblasts within the testis. A hematoxylin purple nuclear counterstain was also used.
Figure 2. Western blot of beta-actin in 3T3 cell lysate probed with rabbit polyclonal anti-beta-Actin (CT) antibody (Cat# 54590). Goat anti-rabbit IgG (H+L), highly cross-adsorbed, HiLyte Fluor™ 750-labeled secondary antibody (Cat# 61056-05-H750 or 61056-1-H750) was used and the signals detected using an infrared-imaging system (Odyssey, Li-Cor).
Figure 3. Western blot of beta-actin in mouse brain (Lane A) and mouse spleen (Lane B) tissue lysate probed with rabbit polyclonal anti-beta-Actin (CT) antibody (Cat# 54590). Immunoreactive bands at ~42 kDa were detected in the lysates. Goat anti-rabbit IgG (H+L), highly cross-adsorbed, HiLyte Fluor™ 750-labeled secondary antibody (Cat# 61056-05-H750 or 61056-1-H750) was used and the signals detected using an infrared-imaging system (Odyssey, Li-Cor).
