Depsipeptide methodology is the latest powerful synthetic strategy to address peptide aggregation and solubility problems in solid phase peptide synthesis. To achieve the goal, isoacryl dipeptides have been developed as building blocks for the synthesis of depsipeptides, which can subsequently be rearranged to the normal peptide sequences via O-N migration under a mild basic condition.
