Biotin-X nitrilotriacetic acid, tripotassium salt (biotin-X NTA) is an excellent reagent for detecting polyhistidine-containing biomolecules immobilized on membranes.
The NTA moiety chelates a Ni++ ion that is also chelated with His tags. The complex can than be detected using streptavidin-conjugates such as streptavidin-HRP. Biotin-X NTA can be removed from the His-tagged molecule at pH 4.8, allowing the blot to be reprobed.
