Native Incretins
Native incretin and metabolic peptides can be modified
to develop analogs with enhanced properties.
| Peptide | Receptor Target | Effect |
|---|---|---|
| GLP-1 Glucagon-like peptide-1 |
GLP-1 receptor (GLP-1R) | Physiological satiety signaling |
| GIP Glucose-dependent insulinotropic polypeptide |
GIP receptor (GIPR) | Glucose and energy regulation |
| Amylin | Amylin receptor (CTR/RAMP complex) | Satiety and gastric emptying |
| PYY | Y2 receptor (Y2R) | Appetite suppression |
| Ghrelin | GHSR | Hunger signaling |
| Glucagon | GIP receptor (GIPR) | Glucose and energy regulation |
Why develop next-generation analogs?
Development of next-generation incretin peptides is focused on improving patient adherence and efficacy, through reduction of GI-related adverse effects, and removal of injection burden by offering oral delivery formats.
Current strategies include extending peptide half-life to reduce dosing frequency and enabling multi-pathway targeting (GLP-1 + GIP + amylin + glucagon).

Modifications for analog development
Native peptide sequences can be modified through a range of approaches, including amino acid substitution with non-canonical amino acids (ncAAs), specialized building blocks, cyclization and conjugation.
Single or multiple modifications can be incorporated into a sequence at varying positions including the N-terminus, C-terminus or internally based on chemistry and sequence compatibility. Each peptide is assessed for production feasibility prior to price quotation.
