This is a naturally occuring protease inhibitor in which the amino terminal has an acetyl group and the carboxy terminal is chemically different than most peptides and proteins, because of an aldehyde group instead of a carbonyl moiety. It is shown to inhibit serine, threonine and cysteine peptidases/proteinases (cathepsins) chiefly acting as a competitive inhitior. It often is used for inhibition of plasmin, trypsin, papain, kallikrein, cathepsin B, as well as, a novel antagonist of calpain. Leupeptin is being developed as an anti-malarial agent and is being studied for its roles in neurodegenerative processes, hearing loss, gingivitis and cardiac, cellular and muscle function pathologies.
