Activation of the coagulation cascade converts fibrinogen to fibrin, through binding of an epitope in the fibrinogen γ chain (γ377–395) to CD11b/CD18 integrin (Mac-1). In the central nervous system, fibrinogen has been associated to demyelination and microglial activation, while this peptide attenuates microglia activation and suppresses relapsing paralysis in multiple sclerosis (MS). Since blocking fibrinogen–Mac-1 interactions affects the pro-inflammatory but not the pro-coagulant properties of fibrinogen, targeting this gamma-fibrinogen epitope could represent a potential therapeutic strategy for MS and other neuroinflammatory diseases associated with blood-brain barrier disruption and microglia activation.
